Oral contraceptives: Potential reduction in contraceptive effectiveness. CYP2C8 enzyme inhibitors (eg, azole antifungals [e.g. ketoconazole] fluvoxamine, gemfibrozil, trimethoprim) Pioglitazone levels in plasma are elevated, and pharmacokinetic effects and adverse reactions are increased. Pioglitazone increases levels of: Amidron, amphetamine, fluoxetine, dextromborphan, mirtazepine, nefazodone, paclitaxel, paroxetine, repliglinin, rosiglitazone. CYP2C8 inducers (phenytoin, phenobarbital, rifampin) reduce the plasma concentrations of pioglitazone, and reduce efficacy.
